Ponatinix is indicated for adults with chronic, accelerated or blast phase chronic myeloid leukemia (CML), or Philadelphia chromosome positive acute lymphoblastic leukemia (Ph+ ALL), that is resistant or intolerant to prior tyrosine kinase inhibitor therapy. It is not recommended for newly diagnosed chronic-phase CML.

The usual starting dose is 45 mg once daily, swallowed whole, with or without food. Your doctor may reduce the dose to 30 mg or 15 mg depending on your response and side effects. Always take it exactly as prescribed.

Ponatinix 15 mg (60 tablets per box) and Ponatinix 45 mg (30 tablets per box), both film-coated tablets in an HDPE pot.

Seek medical help urgently for chest pain, shortness of breath, weakness or numbness, trouble speaking, vision changes, severe headache, unusual bleeding, yellowing of the skin or eyes, or severe stomach pain. Ponatinix can cause serious blood clots, heart problems, liver problems, high blood pressure and pancreatitis.

Your doctor will typically check blood counts every 2 weeks for the first 3 months, lipase every 2 weeks for the first 2 months, liver tests at least monthly, blood pressure regularly, and your eyes at the start and periodically.

Tell your doctor about all medicines and supplements. Some antifungals, antibiotics and antivirals — and grapefruit juice — can increase ponatinib levels, while drugs such as rifampin, carbamazepine, phenytoin and St. John’s Wort can lower them.
